Core Facilities Located in the Sanford Consortium for Regenerative Medicine

There are four shared research cores in the Sanford Consortium building. The core facilities offer investigators access to advanced scientific instruments and services. Such shared technology empowers scientists to better understand and harness the regenerative power of stem cells. These core resources are available on a recharge basis, not only to the resident scientists of the “Collaboratory,” but also to investigators at the participating Sanford Consortium institutions and other research organizations.

Stem Cell Genomics Core

The Stem Cell Genomics Core is a self-service and full-service genomics facility. State of the art equipment as well as guidance are provided to promote the advancement of genomics research and increase level of control and expertise in researchers’ projects. As an example, the novel technology of digital multiplexed gene expression analysis using the NanoString nCounter Sprint Profiler system is available at the core. In addition, the Fluidigm BioMark HD and C1 Single-Cell Auto Prep systems for high gene expression sensitivity, single-cell preparation and analysis are also available as well as the illumina MiSeq sequencing platform to measure genetic variation and gene expression.

Human Embryonic Stem Cell Core Facility

This core provides training for scientists in the basic techniques to work with human embryonic stem cells (hESCs) and other stem cells, providing laboratory space, expertise, and reagents to culture and conduct research with stem cells and their differentiated derivatives. The core also has extensive instrumentation to characterize stem cells and their derivatives, including flow cytometry instruments (both analytical and cell sorting), microscopes (bright-field, fluorescence, and confocal), and high throughput assays (liquid handler, HT-QPCR, multi-label plate reader, automated scanning microscope).

Gene Transfer Targeting & Therapeutics Core 

This core facility provides design, consultation and production services for retrovirus (MMLV/MSCV), lentivirus (HIV/EIAV), adeno-associated virus (AAV), adenovirus (Ad5), rabies virus (g-deleted SAD B19) and vesicular stomatitis virus (VSV) -based viral vector systems. The core offers stocks of DNA shuffled AAV libraries, which allow for selection of novel serotypes with unique transduction properties on cells of interest, as well as non-viral delivery technologies based on mini-intronic plasmid (MIP). In addition to viral/non-viral delivery systems, GT3 offers vector titration services, vector purification services, Replication Competent Lentivirus (RCL) testing, and limited custom cloning and vector services. Several advanced instruments are available for use in the core on a charge-back basis.

Histology and Imaging Core

Provides routine and experimental histological services as well as training and access to various equipment and techniques necessary for the pathological analysis and characterization of tissues and cells.  Histology services include tissue processing and paraffin embedding, H&E and special stains, frozen sectioning, IHC (both manual and automated), thick sectioning for free-floating IHC staining, multi label immunofluorescence staining. Histology equipment available for researchers (following training) includes 2 cryostats for sectioning of thin frozen sections, a vibratome for thick slicing of live tissues, 2 automated rotary microtomes and a sliding microtome for thick slices of fixed tissue. Imaging equipment in the core available to trained users on a chargeback-basis includes a Nikon 80i fluorescence microscope equipped for either color or b&w imaging; an Olympus FluoView1000 Multi-photon confocal microscope on an upright motorized platform with 405, 488, 561 and 536nm laser lines, multi-photon excitation capabilities (including dispersion compensation), and a fast xy-scanner for photo-stimulation experiments.  In addition, there is a Zeiss LSM 780 / Spinning Disk combination microscope on an inverted motorized platform with 405, Argon, 561 and 633nm laser lines, high-sensitivity detection/imaing and full live cell incubation for long-term imaging.

Molecular Imaging Center at Sanford

The Molecular Imaging Center facility supports in vivo imaging with instrumentation including a Bruker 11.7 T MRI, Siemens PET/ SPECT/CT, Vevo 2100 ultrasound, and IVIS Spectrum for imaging luminescence or fluorescence. The imaging suite is set up to enable samples from outside investigators to be analyzed.
